At the beginning of the series, Lisbon maintains a strictly professional relationship with her subordinates, rarely discussing details of her personal life or anything beyond the task at hand. Although Lisbon likes to adhere to proper procedures, Jane constantly clashes with her plans and sometimes directly undermines her authority, often apologizing afterward. While she criticizes most of Jane's antics and revelations in front of him, she often protects him from suspects (especially those with political influence) or from her superiors.
One of the darker conflicts between Lisbon and Jane arises when "Red John," a serial killer who murdered Jane's family, deliberately appears within their jurisdiction. Lisbon is forced to draw a line when Jane reveals that he plans to kill Red John when he is found, putting them at odds (Lisbon wants Red John to be brought to justice, not killed immediately). Lisbon even confronts Jane at the end of the first season, telling him, "Don't you see there are people who care about you, who need you?" hinting at his self-destructive tendencies and dangerous plans involving Red John.
One of Jane's fears is that Lisbon might become a target of Red John at some point because of her close relationship with Jane. Red John once tested their relationship by ordering Jane to bring Lisbon's dead body to him as a "gift."
Lisbon was injured during the Red John investigation. In the final episode of the third season (Strawberries and Cream), Lisbon is shot in the shoulder by Craig O'Laughlin, Van Pelt's fiancé.
Although Lisbon is often frustrated with Jane's behavior, she trusts and supports him. In the final episode of the fourth season (The Crimson Hat), Jane hugs her and says, "Good luck, Teresa. I love you," before faking his death. Later, when she asks him about it, he claims not to remember what he said. At the end of the episode, Jane reaches for her hand.
In the final episode of the fifth season (Red John's Rules), Lisbon is the only person, besides Jane, who knows the names of the seven final suspects of being Red John (Bret Stiles, Gale Bertram, Ray Haffner, Reede Smith, Bob Kirkland, Thomas McAllister, and Brett Partridge). However, in the first episode of Season 6, she asks Van Pelt to track the cell phones of all seven suspects so she can monitor their activities. At the end of the first episode of the season, she receives an urgent call leading her to Brett Partridge's location. Upon arrival, she finds him severely injured, most likely by Red John, uttering his last two words before dying ("Tiger, Tiger"). After Patrick tries calling her multiple times, someone finally answers. To his shock, it is not Lisbon, but his arch-nemesis, who tells him that "Teresa can't come to the phone right now" while cleaning his usual murder weapon. The next thing you see is Red John's hand drawing a smiley face on Lisbon's face, who is now unconscious. But in the second episode, Teresa returns to the CBI after a few days out of the hospital, after Ray Haffner comes to talk. She then tells Van Pelt to stop the investigation of the suspects because it is too dangerous for the CBI.
After the Blake Association scandal, Gale Bertram's crimes, and his connection to Red John, the CBI is disbanded, and Lisbon is detained along with her team while Jane kills the serial killer.

However, after a few months, she becomes the chief of police in Cannon River, a small town in Washington state (My Blue Heaven).
She quits her job as chief to work with Jane for the FBI (Green Thumb).
